You will have so much fun at Martha! She's so fun to watch in between segments during the commercial break. That woman does not play around.
Definitely head to The Redhead for some good Southern food. I love the Low Country shrimp and grits. There are endless places to recommend, but above all, I suggest soaking up the every day life of the city. The tourist stuff is ok, but the what's so special about the city is when you just take it all in and don't try to "do" too much!

You will love the Martha Show! She is INTENSE off camera! Be prepared to lug home pounds of freebies. :)
I moved to NYC from the Bay Area last summer and a couple of my favorites are Saturday brunch at Harry's Steakhouse (near Wall St.) for bottomless champagne cocktails and great food, Fish's Eddy (FUN kitchen store), and Spitzer's Corner in the LES for great beer selection and fancy bar food. :)

I would suggest sitting on the Met's steps, walking through Soho and enjoying an outdoor lunch, and hitting up Uniqlo- it's like Japanese version of H&M but less trendy.

Fun!!! I would head over to ACE hotel for a drink or dinner for sure. And check out the ABC home store. It's like Anthro's home selection but 4 floors of greatness.
Which show are you going to? Are you interested in going to any super-touristy spots? That's pretty much all Jose and I did when we were there, and it was great :) Greenwich Village is the cutest, go see Carrie's "stoop" and to Magnolia Bakery. Central Park is cool too, I recommend Strawberry Fields, and you can see the Dakota (hotel were John Lennon was shot) while you're there. Oh, and also FAO schwartz is fun, haha :P
oh yes i see that someone mentioned fishs eddy...you would might love fishs eddy. and the brooklyn flea! and of course any of the momofuku places!

When are you going? We'll be there in June to see Conan. I didn't know Martha taped in NYC (I never really thought about it) I wonder if I can drag Alex along if I can get tickets. I am totally coming back to this comments section to steal the tips people give you.
